Role of methylation in Pacific oyster response to environmental change

As ocean acidification rapidly impacts marine ecosystems, phenotypic 
plasticity may allow organisms to withstand stressful conditions. 
Genomic methods like epigenetic profiling can elucidate molecular 
mechanisms that contribute to plasticity, allowing for a deep 
understanding of how physiology will be impacted by low pH. In this 
talk, I will explore the role of DNA methylation in Pacific oyster 
(Crassostrea gigas) stress response. I investigated the influence of low 
pH on somatic and reproductive tissue methylomes in adult C. gigas. 
Differential methylation was associated with processes commonly observed 
in oysters exposed to ocean acidification, including apoptosis and 
protein ubiquitination. In gonad tissue, differentially methylated loci 
were also associated with oocyte development. As there was no difference 
in maturation stage between treatments, this finding suggests that 
methylation may maintain gonad growth in low pH. However, adult low pH 
exposure did affect larval survival. Even though oysters spent four 
months in ambient pH conditions between low pH exposure and strip 
spawning, larvae from females that experienced low pH had significantly 
higher mortality. I propose that methylation promotes homeostasis in 
acidified conditions by controlling spurious transcription. While 
transcriptional control could benefit adults, it may be maladaptive for 
larvae when there is a mismatch in parent and offspring environments.
